簡短描述:本15d-PGJ2競爭性ELISA是一種體外酶聯(lián)免疫吸附試驗,用于檢測所有血清、血漿及其他生物流體中的樣本。
背景:本ELISA試劑盒采用競爭性酶聯(lián)免疫吸附試驗(ELISA)原理。本試劑盒提供的微量酶標板已預先包被15d-PGJ2。反應(yīng)過程中,樣本或標準品中的15d-PGJ2與固相載體上固定量的15d-PGJ2競爭結(jié)合針對15d-PGJ2的生物素標記檢測抗體的結(jié)合位點。過量的結(jié)合物及未結(jié)合的樣本或標準品從酶標板上洗去,隨后向每個微孔中加入抗生物素蛋白-辣根過氧化物酶(HRP)偶聯(lián)物并孵育。隨后向每個孔中加入TMB底物溶液。通過加入終止液終止酶-底物反應(yīng),并在450 nm±2 nm波長下通過分光光度法測定顏色變化。通過與標準曲線比較樣品的光密度值(OD),即可測定樣品中15d-PGJ2的濃度。

蛋白胨是一種噴霧干燥粉末,通過動物組織的可控酶解制成。酶包括胃蛋白酶、木瓜蛋白酶和胰酶(后者含胰蛋白酶)。
胃蛋白酶會在含有苯丙氨酸或亮氨酸鍵的任意位置切割肽鏈。
木瓜蛋白酶會在鄰近精氨酸、賴氨酸、苯丙氨酸和甘氨酸的位置切割肽鏈;胰酶則在精氨酸、賴氨酸、酪氨酸、色氨酸、苯丙氨酸和亮氨酸鍵處發(fā)揮作用。
通過水解組織可制得稻草色的蛋白胨,其營養(yǎng)豐富且易溶于水。蛋白胨含有肽類、游離氨基酸和生長因子的混合物。
由于氯化鈉濃度較低,F(xiàn)ormedium™蛋白胨非常適合酵母細胞培養(yǎng)。

組織特異性 :在骨中高度表達。在關(guān)節(jié)軟骨和軟骨細胞中大量表達,且在骨關(guān)節(jié)炎(OA)中表達升高。在關(guān)節(jié)軟骨OA病變的軟骨細胞中與ASPN共定位。
翻譯后修飾 :轉(zhuǎn)化生長因子β1前體蛋白:該前體蛋白在高爾基體內(nèi)被弗林蛋白酶(FURIN)切割,形成轉(zhuǎn)化生長因子β1(TGF-β-1)和潛伏相關(guān)肽(LAP)鏈,二者通過非共價鍵連接,使TGF-β-1處于失活狀態(tài)。潛伏相關(guān)肽:經(jīng)N-糖基化修飾。去糖基化可激活轉(zhuǎn)化生長因子β1(TGF-β-1)。然而,觸發(fā)TGF-β-1去糖基化激活的機制尚不清楚。
功能 :轉(zhuǎn)化生長因子β1前體蛋白:是潛伏相關(guān)肽(LAP)和轉(zhuǎn)化生長因子β1(TGF-β-1)鏈的前體,二者分別構(gòu)成TGF-β-1的調(diào)節(jié)亞基和活性亞基。潛伏相關(guān)肽:在TGF-β-1儲存于細胞外基質(zhì)期間,維持其活性亞基處于潛伏狀態(tài)。通過與TGF-β-1非共價結(jié)合,并與LTBP1、LRRC32/GARP和LRRC33/NRROS等“微環(huán)境分子"相互作用調(diào)控其激活(這些分子控制TGF-β-1的激活)。與LRRC33/NRROS的相互作用可能調(diào)控巨噬細胞和小膠質(zhì)細胞中TGF-β-1的激活。與LRRC32/GARP的相互作用控制活化調(diào)節(jié)性T細胞(Tregs)表面TGF-β-1的激活。與整合素(ITGAV:ITGB6或ITGAV:ITGB8)結(jié)合會導致潛伏相關(guān)肽鏈構(gòu)象改變,進而釋放活性TGF-β-1。
